<p>Method of determining an analyte in a fluid sample, such as whole blood, urine, or saliva, comprising: providing a metal colloid; passing the fluid sample over a surface of a microfluidic channel having disposed thereon a binding partner for the analyte to form an analyte-binding partner complex having the metal colloid associated therewith; flowing a metal precursor solution, such as a silver solution, over the surface to form a metallic layer; flowing in series a predetermined sequence of fluid plugs through the microchannel including first, second and third fluids, wherein the first and second fluids, such as the metal precursor solution and a rinse solution, are separated by the third fluid, for example air, which is immiscible with the first and second fluids; and determining the metallic layer by determining the transmission of light through the layer.</p> |
