Provided is a method for detecting a nucleic acid with which it is possible to inhibit separation of the target nucleic acid that has been hybridized with a capture probe from a substrate during the process of labeling by post-staining and to detect the target nucleic acid at a concentration that is lower than with the sodium ion method but at the same or higher sensitivity. The method for detecting the target nucleic acid comprises (1) a step for forming a double stranded nucleic acid by hybridizing the capture probe and target nucleic acid, (2) a step for bringing the resulting double stranded nucleic acid into contact with a solution containing a label and a divalent metal cation at a concentration of 10 mM or higher in order to introduce the label to the double stranded nucleic acid, and (3) a step for detecting the label that has been introduced into the double stranded nucleic acid.
