PROBLEM TO BE SOLVED: To provide: a carrier onto which Protein A has been immobilized, the Protein A having a specific amino acid sequence enabling the dissociation of antibodies under mild pH conditions (specifically, pH 4.0-5.5) in which many antibodies do not undergo acid modification; and a production method of the carrier.SOLUTION: There is disclosed an immobilization carrier (excluding an immobilization carrier having a monolith structure) onto which a protein is adsorbed by electrostatic interaction. The protein comprises an amino acid sequence represented by general formula R1-R2-R3-R4-R5-R6 and the section of the protein represented by R1-R2-R3 is used for immobilization on the immobilization carrier. In the formula, the sequence represents a sequence from the amino terminal side towards the carboxy terminal side; the sequence of the R2 part is either a protein A mutant sequence, which is the target protein to be immobilized, or a sequence in which 1-3 pieces of the sequences are linked, and the protein A mutant exhibits the characteristics of strongly binding to the antibodies in neutral conditions, and dissociating from the antibodies, which were bonded with in the neutral conditions, in mildly acidic conditions in the range of pH 4.0-5.5. |
