Systems, constructs, and methods for reprogramming cells and loading viral particles are provided. In one aspect, for example, a transformation construct for generating iPS cells can include a vector backbone having a plurality of reprogramming factors, each reprogramming factor being under control of a separate promoter, wherein the vector backbone is contained in an adenovirus particle. In one aspect, the vector backbone has a size of at least 8 kb. In another aspect, the vector backbone has a size of at least 10 kb.
