PROBLEM TO BE SOLVED: To provide a method for discriminating expressed mRNA, enabling different tag discrimination for each specimen in an mRNA expression analysis by monomolecular measurement without influencing on the hybridizing efficiency of a target mRNA when the target mRNA to perform the expression analysis is derived from two or more specimens. SOLUTION: The method for discriminating expressed mRNA includes a step of hybridizing two or more double-stranded DNA complexes having at least one restriction enzyme recognition sequence formed of the first oligonucleotide 101 and the second oligonucleotide 102 having an oligo-dT sequence to a target mRNA of each specimen by the oligo-dT sequence, wherein the restriction enzyme recognition sequences are different from each other in a methylation modification manner.
