The present invention provides a method for preparing dissolved salivary gland tissue, capable of generating stem cells from salivary glands. The method comprises the steps of: adding collagenase to human salivary gland tissue, followed by shake culture, to dissolve the tissue; homogenizing the dissolved tissue; and adding collagenase and hyaluronidase-1 to the homogenized tissue, followed by shake culture, to hydrolyze glycoside bonds in the tissue. Further, the present invention provides a method for preparing adult stem cells derived from human salivary gland tissue, comprising the step of: adding the dissolved salivary gland tissue, which is prepared by the foregoing method, to a medium containing dexamethasone, mercaptoethanol, oncostatin M, and leukemia inhibitory factor (LIF), followed by culture, and then collecting stem cells from the culture liquid. Further, the present invention relates to adult stem cells derived from human salivary gland tissue, wherein the adult stem cells are prepared by the foregoing method and expresses stem cell type cell surface markers CD44, CD49f, CD90, CD105, Oct-4, and Nestin. Further, the present invention relates to a cell therapeutic agent for treating the damage of salivary glands, comprising, as an active ingredient, the adult stem cells derived from human salivary gland tissue. |
