The present invention provides: an optical biosensor comprising a complex comprising a porous support, and an enzyme complex supported inside the pores of the porous support; and a preparation method thereof. A dye does not flow and is concentrated in one place, and thus the sensitivity of the optical biosensor of the present invention is remarkably improved. In addition, it is possible to carry out a quantitative determination and a qualitative analysis since color intensity increases with time.
