The present invention relates to a Mycoplasma contamination confirmation method using the TaqMan real-time PCR method and, more particularly, the TaqMan real-time PCR method using a primer set shown as the base sequence of sequence numbers 1 and 2 and/or a probe shown as the base sequence of sequence number 9. The optimized TaqMan real-time PCR method according to the present invention is useful for specifically detecting broad Mycoplasma. The TaqMan real-time PCR method can be substituted for a negative test method for Mycoplasma by the rapid and accurate detection of Mycoplasma contamination prior to biomedicine production.
