The present invention relates to a biosensor comprising a reduced graphene oxide layer. The biosensor of a horizontal structure includes a reduced graphene oxide layer formed on a substrate, a molecule linker formed on the reduced graphene oxide layer, and a metal nanoparticle layer formed on the molecule linker. The reduced graphene oxide layer has a film structure of one or more layers.
