A method for revealing pathogens in pileate mushrooms (Basidiomicetes), in which, while preparing a preparation of mycelium, a layer of the latter of 2-4 mm is mixed with phosphoric-tungstic acid (PhTA) at pH 5,5-6,5, and the prepared mixture is contrasted with an exposure of 150-180 seconds, coated onto a liner net, dried and photographed on an electron microscope. An assessment by the outward signs is carried out on the basis of images of the contrasted mycelium preparation; and, while compounding a preparation of the fruit body of the mushroom, a sample is taken from a pileus of the fruit body of the mushroom with further contrasting thereof in the PhTA at pH 5,5-6,5 and an exposure of 90-180 seconds, coating the mixture onto the liner net it is dried, and the final assessment of the preparation by the outward signs is carried out on the basis of images thereof. In doing so such structural-morphological objects as bacteria, microscopic fungi and viruses are revealed in integrated manner in one preparation and for short time; in addition bacteria, phages thereof (viruses), microscopic fungi and fragments thereof (organelles) in the preparations of mycelium (fruit body) of the pileate mushrooms (Basidiomicetes) are revealed and fixed on images with magnification by 10000-25000 times, and viruses of a rod-like, bacillus and isometric shape are revealed and photographed at magnification by 25000-45000 times. |
