PROBLEM TO BE SOLVED: To provide isolated monoclonal antibodies that specifically bind to CXCR4 with high affinity, particularly human monoclonal antibodies.SOLUTION: Isolated human monoclonal antibodies bind to human CXCR4 and have the ability to bind to native human CXCR4 expressed on a cell surface, the ability to inhibit SDF-1 binding to human CXCR4, the ability to inhibit SDF-1-induced calcium flux in cells expressing human CXCR4, the ability to inhibit SDF-1-induced migration of cells expressing human CXCR4, the ability to inhibit capillary tube formation by human umbilical vein endothelial cells, the ability to induce apoptosis in cells expressing human CXCR4, the ability to inhibit growth of CXCR4tumor cells. |
