The present invention relates to a three-dimensional cell culture structure with microholes with adhesion onto an agarose gel phase, a method for manufacturing the same, and a three-dimensional cell culture method by attaching cells onto the three-dimensional cell culture structure and culturing the structure by turning the structure upside down. Accordingly, the present invention provides an environment for three-dimensional culture by facilitating a new medium supply and gas circulation without an artificial structure which functions as a crosslinker same as an existing structure and an intracellular structure.
