PURPOSE: A high performance liquid chromatography analysis method of sphingomyelin by enzymatic hydrolysis is provided to accurately analyze dihydrosphingomyelin with reproducibility using an internal standard material. CONSTITUTION: A method for quantitative analysis of an organism-derived sphingomyelin comprises the steps of: adding dihydrosphingomyelin to a biological sample; extracting lipid ingredients from the biological sample; isolating sphingomyelin from the isolated lipid ingredients by high performance thin-layer chromatography (HPTLC); performing enzymatic hydrolysis of isolated sphingomyelin; performing fluorescence derivatization of the enzymatic hydrolysate; and analyzing the reactant by high performance liquid chromatography. |
