PROBLEM TO BE SOLVED: To provide a control method of a device capable of simply constituting an optical detection system in a microchip type microparticle separation device.SOLUTION: There is provided a control method of a microparticle separation device including a step of changing distance between a microchip in which a flow channel through which fluid is conducted, and an orifice for generating a fluid stream at one end of the flow channel are formed, and an objective lens which condenses and irradiates laser to an upstream part of the orifice of the flow channel. The control method includes the steps of: generating the fluid stream; detecting the generated fluid stream; and changing a relative position between the microchip and the objective lens to move the objective lens to a focal position to the microchip after these procedures.
