PROBLEM TO BE SOLVED: To provide a method for producing an oligonucleotide having a phosphodiester structure and phosphotriester structure, applicable also to an oligonucleotide automatic synthesizer.SOLUTION: An oligonucleotide synthesizing method using phosphoamidide technique comprises the following procedure: at least two lineages of phosphoamidide compounds differing in protecting group bound to a phosphite site are used; as one lineage of phosphoamidide compound, such a compound with a phosphite protecting group being a substituent having a dioxolenone skeleton; thereby, in the iodine oxidation step in the procedure of synthesizing the oligonucleotide by the phosphoamidide technique, only the phosphoric acid derived from the lineage of phosphoamidide compound is deprotected into a phosphodiester structure, while the other lineage of phosphoric acid derived from phosphoamidide compound is not deprotected and turned into a phosphotriester structure. |
