PURPOSE: A manufacturing method of polymer microspheres is provided to manufacture the polymer microspheres with an excellent monodispersion property by minimizing the use of sample through the simple method. CONSTITUTION: A manufacturing method of a microsphere by using a polymer monomer and a solvent which is not be mixed with the polymer monomer comprises the following steps. A step of preparing a reproduced mold in which a microwell which satisfies the condition in which a height/maximum width is less than 1.0 and a minimum width/maximum width to the same direction is less than 0.4 is formed. A step of filling a polymer monomer in the microwell of the reproduced mold. A step of applying a solvent which has lower density than the polymer monomer and which has a higher wettability to the reproduced mold than the polymer monomer. A step of polymerizing the polymer monomer.
