A method for evaluation of course of disease at chronic myeloid leukemia includes preparation of suspension of patient’s marrow cells, its cultivation in vitro in nutrient medium RPMI-1640 and analysis of morpho-functional properties of cultivated cells. Additionally analysis of proliferative potential of cell-precursors of suspension of patient’s marrow cells is performed, that for fraction of mononuclear cells is separated from it. Total number of obtained cells and percent of viable cells in suspension are calculated with use of dye for selective dyeing dead cells. Cellularity of suspension is brought to needed concentration, this is determined empirically, with its cultivation in semi-fluid agar with addition of granulocyte-macrophage colony-stimulating factor during not less than fourteen days. After that by means of inverted microscope number of cell aggregations formed as result of cultivation is calculated. At following statistical processing of the results obtained the level of proliferative potential as ratio between granulocyte colonies and clusters is determined, with making conclusion on occurrence of remission of the disease. |
