The invention relates to the field of biochemistry and concerns a method for inhibiting the activity of enzymes of nucleic acid biosynthesis, which are responsible for forming DNA macromolecules and ribonucleic acid (RNA) macromolecules complementary to the initial template DNA in a cell of a living organism. In particular, the invention concerns a method for inhibiting the activity of the enzyme RNA polymerase, which is responsible for carrying out the transcription of deoxyribonucleic acid (DNA). This invention can be used, for example, in molecular biology, biochemistry, pharmacology, etc. The invention extends the range of technical agents which can be used for inhibiting the activity of enzymes of nucleic acid biosynthesis, in particular for inhibiting the activity of the enzyme RNA polymerase. This is achieved in that the method for inhibiting the activity of enzymes of nucleic acid biosynthesis, in particular for inhibiting the activity of the enzyme RNA polymerase, by administering an inhibitor on the basis of a compound containing an organic, inorganic or hybrid macrocyclic fragment to the transcription system, is characterized in that the inhibitor used is at least one macropolycyclic compound, in particular, a macropolycyclic complex, with an encapsulated metal ion, and also in that said compound blocks the macromolecular-complex interface between the enzyme, the template nucleic acid and the resulting polynucleotide.
