<p>The present invention provides a method, kit and system for diagnosis of leukemia. The method comprises the following steps: extracting whole blood DNA of a testee, dissolving the DNA in a buffer solution with the pH value being 6~8 to obtain a DNA solution A; heating the solution A at the temperature of 80°C~95°C for 2 to 15 minutes, then performing fast quenching on the solution A at a temperature below 20°C, and placing the solution still overnight at the quenching temperature to obtain a solution B; mixing a sufficient quantity of the solution B and a sufficient quantity of the buffer solution with the pH value being 6~8 and cyanine dye to prepare a solution C; measuring the following indexes of the solution C separately: the highest absorbance of the absorption peak within the range of 550-560 nm, marked as AM; the highest absorbance of the absorption peak within the range of 515-525 nm, marked as AD; the highest fluorescence intensity of the fluorescent emission peak within the range of 570-580 nm, marked as FlM; the highest fluorescence intensity of the fluorescent emission peak within the range of 605-615 nm, marked as FlD; calculating MD (A) = AM/AD and MD(Fl)=FlM/FlD according to the above measurement results; and if MD(A) is smaller than 1.4, or MD(Fl) is smaller than 5.2, confirming that the testee is afflicted with leukemia.</p> |
