A method for determination of activity of gelatinases in human blood plasma includes electrophoresis of preliminarily diluted in Laemmli's buffer test samples in gelatin-polyacrylamide gel with sodium dodecylsulphate, gel washing with Triton X-100, incubation in enzyme buffer, dyeing in Coomassie solution G250, decoloration with solution of methanolacetic acid – water, densitometry of gel and calculation of amount of gelatinases with respect to standard sample. At that the samples of blood plasma are preliminarily incubated, diluted in buffered physiological solution, with adding Laemmli's buffer, with electrophoresis in polyacrylamide gel with following transformation of treated gel to digital format, and with calculation of activity of gelatinases by means of the program Sorbfil Videodensitometer 2.0, with use of the blood plasma pool from 20 healthy donors, activity of gelatinases in which is taken to be 100 %, as a reference standard. |
