PROBLEM TO BE SOLVED: To provide a method for quantifying an amino acid in a biological sample without being affected by a great amount of various impurities such as inorganic phosphoric acids and urea included in the biological sample.SOLUTION: An enzyme using a target amino acid as a substrate and generating a pyrophosphate from an adenosine triphosphate (ATP) is used. The generated pyrophosphate is reacted with a pyruvate phosphate dikinase, and the amino acid is quantified based on the amount of generated pyruvic acid. Specifically, methionine-selective quantification can be carried out without performing any pretreatment by reacting an adenosylmethionine synthetase with methionine and measuring generated pyrophosphate with the pyrophosphate quantification system. Alternatively, citrulline can be quantified by reacting an argininosuccinate synthetase with citrulline and measuring generated pyrophosphate with the pyrophosphate quantification system. Furthermore, when arginine deiminase is reacted with arginine and generated citrulline is measured with the citrulline quantification system, an amount of the arginine can be determined. |
