FIELD: veterinary science.SUBSTANCE: method to diagnose chlamydia in breeding boars includes sperm survey and detection of reticular chlamydia corpuscles in it by the method of electronic microscopy. The produced sperm concentrate by means of centrifuging is fixed with 2.5% glutaraldehyde, washed in phosphate buffer, treated with osmic fixator, poured into araldite and epon of identical firms. Cuts are produced on the ultratome LKB-III, concentrated with 2% alcohol solution of uranyl acetate and lead citrate by Reynolds, viewed in the electronic microscope of EMV-100BR type with accelerating voltage 75K, searching for and indication of affected areas is done by the method of target ultramicrotomy for production of semi-thin cuts and their dyeing with toluidine blue as instructed by A.I.Lysenko.EFFECT: method improvement.1 tbl, 3 dwg |
