PURPOSE: An aptamer screening method using graphene and a Nampt-specific aptamer are provided to remove single stranded DNA which is not bound to a target. CONSTITUTION: A method for preparing a DNA aptamer of a target non-immobilization type comprises a step of removing a single stranded DNA with 30-50 bases from a mixture of a single stranded DNA pool and a target material using a graphene. The method further comprises a step of repeating a graphene-based selection and a counter selection.
