PROBLEM TO BE SOLVED: To provide a new useful drug-resistant marker gene in a microorganism of the genus Aspergillus.
SOLUTION: It is found that the microorganism belonging to the genus Aspergillus becomes sensitive to carboxin by culturing the microorganism in a culture medium containing acetic acid as a carbon source. Furthermore, it is predicted that a carboxin-resistant microorganism appears and the carboxin-resistant mutant gene is produced by performing a mutation-inducing treatment of the gene of the microorganism belonging to the genus Aspergillus. In this way, a gene corresponding to a succinate dehydrogenase Ip subunit gene known as a carboxin-resistant gene of a Basidiomycetes is searched from the whole base sequences of Aspergillus oryzae RIB40. As a result, it is confirmed that the mutation is caused in the gene.
COPYRIGHT: (C)2009,JPO&INPIT |
