PURPOSE: A method and a kit for discriminating Oreorchis patens and Oreorchis coreana Finet are provided to easily discriminate the Oreorchis patens and the Oreorchis coreana Finet by using a direct sequencing method, an SNP-genotyping method, and an RFLP(Restriction fragment length polymorphism) method. CONSTITUTION: A method for discriminating Oreorchis patens and Oreorchis coreana Finet comprises: a step of preparing biological samples from the Oreorchis patens and the Oreorchis coreana Finet; a step of isolating genome DNA from the biological samples; a step of amplifying a DNA sequence containing a GCCGGC sequence, which is a recognition site of a Nael restriction enzyme in an ITS sequence of the Oreorchis coreana Finet; a step of amplifying a DNA sequence containing a GCNGGC sequence, which is an alternative sequence of the Oreorchis patens; and a step of treating the amplified product with NaeI. |
