PROBLEM TO BE SOLVED: To provide a method for semiquantitatively or quantitatively analyzing a ratio between a cysteine residue and a formyl glycine residue in protein material.SOLUTION: A method for analyzing a ratio between a cysteine residue and a formyl glycine residue in protein material includes a step (a) for making the protein material into (i) a halogen carboxylic acid label, (ii) a halogen-substituted carboxylic acid amide label, and (iii) a halogen-substituted carboxylic acid label, next, a hydrazine label, or a halogen-substituted carboxylic acid label, and then an oxime formation label, a step (b) for digesting each labeled protein to be each peptide fragment mixture, a step (c) for adding the acquired mixture to reverse phase chromatography to mutually separate peptide fragments, acquiring chromatogram, and a step (d) for specifying a peptide segment containing the cysteine residue and a peak resulting from the peptide segment containing the formyl glycine residue by mutually comparing the chromatogram. |
