PURPOSE: A method for preparing isomalto oligosaccharides is provided to simplify existing complex processes without steps of pulverizing and steaming grains, and to produce a large amount of isomalto oligosaccharides. CONSTITUTION: A method for preparing isomalto oligosaccharides comprises: a step of adding liquefying enzyme to whole grains and liquefying; a step of adding saccharifying enzyme to the liquefied solution and saccharifying; and a step of adding transferase to the saccharified solution without steps of pulverizing and steaming grains. The whole grains are selected from the group consisting of rice, barley, soybean, millet, hog millet, sorghum, wheat, corn, oat, and buckwheat.
