<p>Patent No. 593703 Disclosed is a method for detection of bactericidal antibodies in a biological sample, the method comprising: combining in a reaction mixture: a biological sample suspected of containing bactericidal antibodies, wherein where the biological sample is from a human subject then said sample was obtained previously from the human subject; a viable Gram-negative pathogenic bacterium of interest; and fresh human whole blood obtained previously from a non-immune human donor which does not contain detectable bactericidal antibodies effective against the pathogenic bacterium of interest, wherein when the biological sample is obtained previously from a human subject, the human from whom the human biological sample was obtained previously is not the non-immune human donor, and wherein said fresh human whole blood contains an anticoagulant that does not significantly affect complement activation or complement activity; and detecting the presence or absence of bactericidal antibodies in said sample by assessing viability of said Gram negative bacterium, wherein decreased viability of said Gram negative bacterium in the presence of said biological sample compared to viability of said Gram negative bacterium in a control not containing bactericidal anti-bacterial antibodies, indicates the sample contains bactericidal anti-bacterial antibodies.</p> |
