<P>PROBLEM TO BE SOLVED: To provide a method of specifically measuring glycolated β-chain N-end of glycolated hemoglobin by using an enzyme without resort to any separation operation, and to provide a measurement reagent kit. <P>SOLUTION: A protease cutting out a glycolated amino acid and/or a glycolated peptide form the glycolated β-chain N-end substantially without cutting out any glycolated amino acid or glycolated peptide from glycolated α-chain N-end of the glycolated hemoglobin or a fragment thereof is screened. By using the protease obtained by the screening method, a method and a measurement reagent kit by which the glycolated β-chain N-end of the glycolated hemoglobin is specifically measured without resort to any separation operation, can be provided.
