Abstract The current invention features methods of enhancing the efficacy and specificity of RNAi. The invention also provides a method of decreasing silencing of an inadvertent target by an RNAi agent. Furthermore, the invention features compositions, including siRNAs, shRNAs, as well as vectors and transgenes for mediating RNAi. The RNAi agents of the invention have improved specificity and efficacy in mediating silencing of a target gene. |
