NOVEL DNA BINDING DOMAIN AND NOVEL DNA BREAKING ENZYME INCLUDING THE SAME

PROBLEM TO BE SOLVED: To provide an artificial DNA binding domain allowing any base sequence to be freely made a target gene and having accurate binding specificity.

SOLUTION: DNA binding protein that specifically recognizes and binds a target sequence contains the following domains in order from an N terminal: (1) a domain with a 250-312 amino acid length; (2-1) a target sequence recognition domain of TALE or a variant target sequence recognition domain of TALE; (2-2) a target sequence recognition final domain of TALE or a variant target sequence recognition final domain of TALE; (3) a domain with a length equivalent to a target sequence comprising 7 to 8 base pairs; and (4) a zinc finger domain or a variant zinc finger domain.
