<p>The antibiotic fusafungine is produced by cultivating Fusarium Lateritium WR, CSB 119.63 in a conventional culture medium therefore by deep culture with aeration at a rate of from 0.7 to 0.9 litres of sterile air per minute per litre of nutrient solution and simultaneous agitation, the pH of the culture medium being maintained within the range of 4.5 to 7.0 by means of one or more buffers. A typical medium is an aqueous solution of saccharose, glucose, ammonium nitrate, sodium chloride, dihydrogen potassium phosphate and magnesium sulphate. Temperature of cultivation is 25 DEG to 33 DEG C. and duration is 60 to 70 hours. The antibiotic is isolated from the mycelium or whole broth by extraction with a solvent such as methanol, methylethylketone, ethyl acetate or hexane. Further purification may be obtained by counter-current extraction of a hydrocarbon solution by a methanol-water mixture, followed by crystallization by the addition of water. Fusafungine has antibacterial and anti-inflammatory properties and may be applied topically in the form of a solution, aerosol or ointment.</p> |
