Provided are method and kit for enriching and purifying stem cells, and uses thereof. The method and kit can be used directly for enriching and purifying stem cells derived from rodent, human or other mammals. With this method and kit, stem cells can be effectively and precisely enriched and purified from said cell population. Moreover, the stem cells obtained from the method and kit can also be without any external marks. The kit has the advantages of simple to use, industrial production, and practical and effective. Also provided are the obtained stem cells and uses thereof such as in research, diagnosis, drug screening or therapy.
