Provided is an activity-enhanced L-asparaginase variant, which is a variant of a bacillus coli wild L-asparaginase shown in SEQ ID NO: 3, and comprises an amino acid sequence having one or more amino acid substitutions at the 48th, 49th, 152nd and 283rd bits corresponding to the SEQ ID NO: 3. Provided are an isolated nucleic acid of a nucleotide sequence comprising the L-asparaginase variant, a recombinant expression construct comprising the nucleic acid, and a recombinant host cell comprising the expression construct. Further provided is a method of producing the L-asparaginase variant and a pharmaceutical composition that comprises the L-asparaginase variant and is used for treatment of tumors.
