PROBLEM TO BE SOLVED: To provide a method for easily preparing a double stranded DNA that has a desired sticky end (protruding end), and a method for preparing a simple and highly versatile recombinant DNA and a vector using a DNA fragment prepared by the method.

SOLUTION: The DNA strand is synthesized using a DNA polymerase having a defective of injury bypass activity (injured base intolerant) and a primer that contains an injured base, and the sticky end (protruding end) is prepared by stopping the DNA polymerase.
