A recombinant spike protein comprising a canine coronavirus neutralization antigen epitope, and a vaccine composition comprising the same protein are provided to prevent or treat bowel disease caused by canine coronavirus in dogs. The recombinant spike protein comprises the canine coronavirus neutralization antigen epitope, wherein the recombinant spike protein has the amino acid sequence selected from SEQ ID NO:23, SEQ ID NO:24, SEQ ID NO:25, SEQ ID NO:26, SEQ ID NO:27 and SEQ ID NO:28. An expression vector contains the gene encoding the recombinant spike protein, wherein the vector is pMAL-c2x. A method for producing the recombinant spike protein comprises the steps of: (a) constructing the recombinant expression vector containing the recombinant spike protein gene and an expression control sequence operatively linked to the protein gene; (b) transforming a host cell with the recombinant expression vector; and (c) culturing the transformed cell in a medium and recovering the spike protein. A yoke antibody against the spike recombinant protein is prepared by injecting the recombinant spike protein into laying hens, recovering eggs from the immunized laying hens, and separating the antibody from the yolk of eggs. |
