PURPOSE: A hybrid silica nanoparticle for intracellular transmission of siRNA is provided to reduce toxicity and to control surface potential. CONSTITUTION: A hybrid silica nanoparticle for intracellular transmission of siRNA is formed in a core-shell form comprising polyethylene imine-polyethylene glycol(PEI-PEG) copolymers on the surface of the silica nanoparticles. A method for preparing the hybrid silica nanoparticles comprises: a step of the PEI-PEG copolymers by chemical reaction of PEI amine and PEG; a step of preparing compound(PEI-PEGsilane) attached to the silica nanoparticles; and a step of dehydrocondensation of the compound(PEI-PEGsaline) and silica nanoparticles. A pharmaceutical composition for suppressing gene expression contains the siRNA complex and pharmaceutically acceptable carrier. |
