PURPOSE: A transgenic allotriploid sterile fluorescent Oryzias dancena with novel fluorescence phenotype is provided to ensure ornamental value without harm to an ecosystem. CONSTITUTION: A fluorescence expression vector expressing fluourescence in whole tissue of Oryzias dancena contains a beta-actin gene promoter and red fluorescence protein(RFP) structure gene. The fluorescence expression vector, podb-actRFP, additionally contains beta-actin 3'-UTR fragment containing polyadenylation signal. A method for producing homozygous transgenic Oryzias dancena(F2) comprises: a step of injecting transforming gene unit of the expression vector into fertilized eggs of Oryzias dancena and incubating to obtain transgenic Oryzias dancena(F0); and a step of crossbreeding the F0 with non-trangenic Oryzias dancena to obtain hemizygous transgenic Oryzias dancena(F1). |
