METHOD FOR MANUFACTURING ACTIVE RECOMBINANT BLOOD COAGULATION FACTOR IX

PURPOSE: An expression vector which prepares recombinant blood coagulation factor IX is provided to enhance recombinant proteins and to maximize recombinant blood coagulation factor IX production. CONSTITUTION: A method for preparing human blood coagulation factor IX comprises a step of culturing mammalian cells in a serum-free medium containing yeast hydrolysate, soybean hydrolysate, or a mixture thereof. The concentration of the yeast hydrolysate is 1-10 g/L. A recombinant expression vector has: human beta-globin MAR element nucleotide sequence and intron 1 fragment. The intron 1 fragment has 141bp-981bp nucleotide sequence with a splicing donor at 5' terminal of intron 1 and 157bp-444bp nucleotide sequence with a splicing receptor at 3'-terminal of intron 1.
