Provided are polynucleotide sequences, which can be used in pyripyropene A synthesis. The nucleotides encode a polypeptide having any one or more activities of polyketide synthase activity, prenyltransferase activity, hydroxylase activity, acetyltransferase activity or adenylate synthetase activity. The polynucleotide can be derived from Penicillium coprobium PF1169 strain and the methods of synthesis using specified precursors as target for the encoded polypeptides are also provided.
