PURPOSE: A sustained release formulation containing donepezil or pharmaceutically acceptable salt thereof is provided to effectively control release speed of donepezil or salt and to improve elusion safety. CONSTITUTION: A sustained release pharmaceutical composition contains donepezil or pharmaceutically acceptable salt thereof, and polyvinyl acetate or fatty acid ester. The salt is donepezil hydrochlorie. The polyvinyl acetate contains polyvinyl pyrrolidone and polyvinyl acetate in a weight ratio of 8:1~2. The fatty acid ester is glyceryl behenate, hydrogenated castor oil, stearyl, cetostearyl or cetyl alcohol, mono-, di, or tri glyceride, or solid paraffin. The composition additionally contains elution controlling agent such as carboxy methyl ethyl cellulose or methacrylic acid-acrylic acid ethyl copolymers. The composition also contains pharmaceutically acceptable additives. The composition is used for preventing and treating Alzheimer's disease.
