PURPOSE: A medium for maintaining the stemness of human embryonic stem cells or dedifferentiation pluripotent stem cells is provided to enable long-term culture(sub-culture). CONSTITUTION: A medium for maintaining the stemness of human embryonic stem cells or dedifferentiation pluripotent stem cells contains: protein kinase C inhibitor as a stemness-maintaining agent; a histone deacetylase inhibitor; or bone morphogenic protein channel blocker in a basic medium. The bone morphogenic protein channel block is noggin, chordin, follistatin, and 6-(4-(2-piperidine-1-ylethoxy)phenyl)-3-pyridine-4-ylpyrazolo(1,5-a)pyrimidine. The basic medium is a xenopathogen-free medium. A method for culturing the human embryonic stem cells or dedifferentiation pluripotent stem cells comprises a step of culturing human embryonic stem cells or dedifferentiation pluripotent stem cells.
