Provided is a pharmaceutical composition comprising (i) an isolated polypeptide comprising a specified amino acid sequence that has reduced procoagulant activity compared to wild-type factor Xa, (c) is capable of binding to a factor Xa inhibitor and (d) does not assemble into a prothrombinase complex, and (ii) a blood coagulating agent. The synergistic combination can be used to treat or prevent bleeding in a patient having anticoagulant therapy with fXa inhibitor.
