<P>PROBLEM TO BE SOLVED: To provide a DNA construct useful for efficient production of hprt gene recombinant cell in dhfr gene-deleted CHO cell. <P>SOLUTION: The DNA construct comprises a first homologous DNA fragment, a target protein gene, and a second homologous DNA fragment from 5' terminal toward 3' terminal, wherein the first homologous DNA fragment and a second homologous DNA fragment have homology which allows recombination with a part of genetic locus of a hypoxanthine-phosphoribosyl transferase enzyme (hprt) of the dhfr gene-deleted CHO cell genome, the chain length is 1kbp or more, and the dhfr gene is used as a selection marker gene. <P>COPYRIGHT: (C)2013,JPO&INPIT |
