<p>[Problem] The objective of the invention is, in a detection method and a detection device for optically measuring an amount of a substance to be detected in a sample liquid by utilizing surface plasmon enhancement effect, to reduce an error caused by fluctuation of the angle of incidence of excitation light relative to a metal film and to increase detection sensitivity. [Solution] A sensor chip, which is provided with a sensor portion formed by disposing a metal film (16) on one surface of a dielectric plate (17), the sensor portion being divided into five planar regions (E1-E5) having different angles of inclination relative to a predetermined reference plane, is used such that: a sample liquid containing a substance to be detected is caused to contact the sensor portion of the sensor chip, whereby a fluorescence-labeled binding substance in an amount corresponding to an amount of the substance to be detected contained in the sample liquid is bound to the sensor portion; excitation light is irradiated onto each of the planar regions (E1-E5) of the sensor chip sequentially; an amount of light generated at the sensor portion in response to irradiation of excitation light is detected for each planar region (E1-E5) of the sensor chip; and an amount of the substance to be detected is measured on the basis of the detection result for one of the planar regions (E1-E5) for which the maximum amount of light has been detected.</p> |
