A tissue engineering scaffold for application to a wound comprises a tissue regeneration matrix layer 1 for contacting the wound and a protective backing element 2 bonded to the tissue regeneration matrix layer. The tissue regeneration matrix layer is formed from a porous biodegradable material which takes up cellular material from the wound and supports tissue generation, whilst the protective backing element is formed from a water-impermeable material. The protective backing element is provided with at least one cavity therein containing an aqueous medium, the cavity being in fluid communication with the tissue regeneration matrix layer so that one or more biomarker substances absorbed by the tissue regeneration matrix layer can diffuse into the aqueous medium within the cavity. To monitor the progress of wound healing the aqueous medium is withdrawn from the cavity for analysis to identify one or more biomarkers in the aqueous medium. Preferably, the backing element is formed from a silicone polymer, whilst the tissue regeneration layer may comprise a collagen or gelatine cryogel. The cavity can be a bubble accessible by a syringe needle to extract the aqueous medium.
