<p>The subject of invention which falls within the scope of tissue engineering is process of establishing a differentiated urothelium in connective tissue of intact human amniotic membrane as a tissue holder and thus the construct of connective tissue of intact amnioticmembrane and the urothelium with a high degree of differentiation is established. According to the procedure for establishing a highly differentiated urothelium, as tissue holder the connective tissue of intact human amniotic membrane is used. The latter, due to high levels of collagens glycoproteins and proteoglycans with bound growth factors and growth factors which are released from the amniotic epithelial cells as well, represent a tissue unique tissue holder which promotes cell differentiation. Urothelial cells in the connective tissue of intact amniotic membrane form urothelium, consisting of basal, intermediate, and highly differentiated surface urothelial cells. Highly differentiated urothelium grown in connective tissue of intact amniotic membrane provides a blood- urine barrier, thus effectively mimics the action of urothelium in vivo. This represents a unique model for both basic and applied studies of lower urinary tract. Established model allows preclinical testing of various substances as well as the development of potential new medicines.</p> |
