Disclosed herein is a method of preparing frozen or lyophilized microbial cellswherein a permeate obtained by filtering milk or whey with ultra-filtration (UF) membrane is mixed with microbial cells, preferably selected from Bifidobacterium, Lactobacillus, Streptococcus and Lactococcus; and the mixture is frozen or lyophilized, wherein prior to mixing the permate with the microbial cells, the lactose in the permeate obtained by filtering milk or whey with ultra-filtration (UF) membrane is decomposed with enzyme and/or heat.
