The invention relates to microbiology and the medical industry and also to genetic engineering and biotechnology. By engineering recombinant plasmid DNA containing the structural gene of human interferon alpha-2b under the control of a regulated promoter, a strain of Escherichia coli bacteria is produced which provides the synthesis of the precursor protein of mature methionine-free human interferon alpha-2b in an insoluble form. In the precursor protein, the sequence of mature methionine-free interferon alpha-2b is fused with the sequence of the SUMO protein (SMT3) of the yeast Saccharomyces cerevisiae. A strain of Escherichia coli bacteria is produced which provides the biosynthesis of ULP275 protease for the enzymatic processing of the precursor protein of methionine-free interferon. A method is proposed for producing methionine-free interferon, which involves the joint implementation of the processes of renaturation, disulphide bond formation and enzymatic isolation of methionine-free interferon alpha-2b from the precursor protein under the effect of ULP275 protease. |
