<P>PROBLEM TO BE SOLVED: To provide a method for the introduction of predetermined genetic changes in target genes of a single-stranded oligodeoxynucleotide, a certain derivative thereof, and a living cell. <P>SOLUTION: The single stranded oligonucleotide encoding a predetermined genetic change is modified by the attachment of 3' and 5' blocking substituents such as a 3'-3' linked cytosine nucleotide and a 5' linked indocarbocyanine dye. The modification can consist of the replacement of the 3' most and/or 5' most intemucleotide phosphodiester linkage with a non-hydrolyzeable linkage such as a phosphorothioatediester linkage or a phosphoramidate linkage. A composition consisting of the oligodeoxynucleotide encoding the genetic change and a macromolecular carrier contains a polycation, an aqueous-cored lipid vesicle, or a lipid nanosphere as the carrier. <P>COPYRIGHT: (C)2012,JPO&INPIT |
